ABSTRACT

Objective:To investigate the surgical management of perifolliculitis capitis abscedenset (PCAS).Methods:From June 2015 to October 2018, 8 patients with PCAS were treated, aged 13-26 years, with an average age of 23 years. Hypepigmented plaques in size were from 0.5-4.0 cm. The operative technique was based on the complete excision of the entire diseased skin and subcutaneous fatty tissue, the fistula was explored using a probe, the seton was inserted through the remaining tract in a double-strand fashion, this double-strand elastic seton was then tied over itself without excessive tension. The wound was covered with absorptive dressing. Results:All the wounds in 8 patients healed primarily. All patients were followed up for 6 months and showed no recurrence after surgery. Conclusions:Perifolliculitis capitis abscedens et suffodiens scalp is a rare chronic suppurative skin disease, its etiology and pathogenesis are unclear, but related with follicular atresia, bacterial infection, immune factors, sex hormones, smoking and obesity. The treatment is difficult, including antibiotics, vitamin A acid, biological agents, photodynamic, laser and surgery. The disease is stubborn and refractory. The technique of wide surgical excision and loose setons drainage is an effective method for PCAS.

ABSTRACT

Objective:To investigate the level of rumination and its influential factors among Chinese psychiatry nurses who go through work place violence. Methods:In this study, 150 psychiatry nurses were recruited from Affiliated Brain Hospital of Guangzhou Medical University (specialize in psychiatry), via the combination of convenient sampling and snowball sampling. Chinese Event-related Rumination Inventory (C-ERRI) was applied in the survey. Results:The total score of C-ERRI was (22.11±9.62) points, and the scores of intrusive rumination subscale and deliberate rumination subscale were (12.99±5.58) and (9.12±6.01) points, respectively. Multiple Linear Regression Analysis show that assault frequency in recent year ( B=-3.195, P<0.01) and whether got injury in the recent assault ( B=8.591, P<0.01) were predictors of deliberate rumination, which account for 26.8% variance of the equation. Gender (male) ( B=-2.415, P<0.01), Injury frequency in recent year ( B=2.864, P<0.01) and whether got injury in the recent assault ( B=8.949, P<0.01) were predictors of intrusive rumination. They account for 36.0% variance of the equation. Conclusion:In this study, the level of rumination among Chinese psychiatry nurses was low. Their rumination style was deliberate rumination.

ABSTRACT

Objective:To study whether aspirin has inhibitory effect on microglia activation induced by Poly-IC and its mechanism. Methods:Microglia cell line BV2 were cultured in vitro to establish a Poly-IC stimulation-induced microglia cell immune activation model. The experiment groups were divided into control group (no treatment), model group (Poly-IC 10 μg/ml), high dose aspirin group (1 mmol/L aspirin), low dose aspirin group (0.1 mmol/L aspirin), high dose aspirin pretreatment group (Poly-IC 10 μg/ml + 1 mmol/L aspirin) and low dose aspirin pretreatment group (Poly-IC 10 μg/ml + 0.1 mmol/L aspirin). The phagocytosis ability of microglia cells, reactive oxygen species (ROS) and Iba1 protein expression were detected by using immunofluorescence method. The expression of the inflammatory cytokines Il-1β, Il-6, Il-10, TNF-α and cox-2 mRNA in microglia cells were detected by real-time quantitative PCR (RT-qPCR).Results:Compared with the control group, the morphology of microglia cells in model group changed significantly, and the phagocytosis ability and production of reactive oxygen species (ROS) increased. At the meantime, the expression of Iba1 protein was strongly decreased. In the model group, The mRNA expressions of IL-1β(20.55±1.92), IL-6 (63.98±7.83), TNF-α (16.84±3.19), COX-2 (6.78±0.42) were higher than IL-1β(1.01±0.14), IL-6 (0.95±0.17), TNF-α (1.22±0.38), COX-2 (0.87±0.11) in the control group. (Il-1β ( t=26.14), Il-6 ( t=10.22), TNF-α ( t=17.06) and COX-2 ( t=37.07), all P<0.01). In the aspirin pretreatment group, the phagocytic ability of microglia cells was inhibited compared with the model group, and the production of reactive oxygen species (ROS) reduced. The expression of Iba1 protein was also partly recovered. Meanwhile, the effect of the high aspirin dose pretreatment group on pro-inflammatory factors IL-1β(9.95±0.52), IL-6 (39.64±6.89), TNF-α(1.57±0.42), COX-2 (2.47±0.14)were lower than those in the model group significantly.(IL-1β: t=14.18, IL-6: t=3.69, TNF-α: t=16.68, COX-2: t=27.03, all P<0.01). Conclusion:Aspirin has an inhibitory effect on microglial activation induced by Poly-IC, which may be related with inhibiting the expression of inflammatory factors.

ABSTRACT

BACKGROUND:Bone marrow mesenchymal stem cel s transplanted into rats exposed to silica dust can home to the injured lung, but the homing effects via different ways are stil unclear. OBJECTIVE:To comparatively observe the distribution of bone marrow mesenchymal stem cel s transplanted via different ways into rats exposed to silica dust. METHODS:Bone marrow mesenchymal stem cel s of donor rats were isolated through whole bone marrow adherent method and transfected by Lv-eGFP. Receptor rats were exposed to silica dust through windpipe injection and randomly divided into intravenous injection and intratracheal injection groups. Then, transfected bone marrow mesenchymal stem cel s were injected via the vein and trachea into acceptor rats. The acceptor rats were kil ed at weeks 1, 2, 3, 4 after transplantation to take the lung, heart, liver, spleen, kidney, and brain tissue that were made into frozen sections and observed under fluorescence microscopy. The intensity of green fluorescence (absorbance value) was analyzed using image analysis software. RESULTS AND CONCLUSION:Strong, wide and lasting green fluorescence was both observed in the lung tissue of intravenous injection and intratracheal injection groups, which was especial y remarkable around the bronchus and blood vessels. The fluorescence intensities of both two groups were slightly decreased with time, but there was no difference between the two groups (P>0.05). The fluorescence in the other organs of both two groups was also observed at early stage. It was stronger and wider in the liver, spleen and heart, while fainter and less in the kidney and brain, and reduced with time in al the organs. Fluorescence could be observed few and faint only in the liver and spleen at late stage, and could hardly be seen in the brain. The fluorescence intensities of the liver, spleen, heart, kidney and brain had no significant difference between the two groups at the same time (P>0.05), but the fluorescence intensity in the brain at the 1st week showed significant difference between the two groups (P<0.05). These findings indicate that the intravenous injection and intratracheal injection of bone marrow mesenchymal stem cel s have similar homing effects in rats exposed to silica dust.

ABSTRACT

BACKGROUND:Bone marrow mesenchymal stem cels transplantationvia the trachea can relieve the lung injury of rats exposed to silica dust, but their distribution and migrationin vivois stil unclear. OBJECTIVE:To investigate the distribution and homing of bone marrow mesenchymal stem cels transplanted via the trachea into rats exposed to silica dust. METHODS: Bone marrow mesenchymal stem cels from Sprague-Dawley rats were isolated through bone marrow adherent method and transfected with lentivirus carrying enhanced green fluorescent protein gene (Lv-eGFP). Trypan blue staining and cel counting kit-8 were applied to assay the viability and proliferation of the transfected and untransfected cels. Sprague-Dawley rats, SPF level, were randomized into control group and silica dust exposure group. Rats in the two groups were respectively injectedvia the trachea with 1 mL of sterile silica dust suspensions (40 g/L) and 1 mL of normal saline. At 2 days after modeling, 2.2×106 transfected bone marrow mesenchymal stem cels were injected via the trachea into the rats of control group and silica dust exposure group. Rats were kiled at weeks 1, 2, 3, 4 after transplantation, and the distribution and intensity of green fluorescence in the lung, heart, liver, spleen, kidney, and brain tissue were observed under the fluorescence microscopy by frozen sections and analyzed using imaging analysis software. RESULTS AND CONCLUSION: When the multiplicity of infection was 50, there were no significant differences between the viability and proliferation activity of the transfected and untransfected cels (P > 0.05). After transplantation of transfected bone marrow mesenchymal stem cels, strong green fluorescence was observed widely in the lung, especialy around the bronchus and blood vessels, and stil obvious at the 4th week. The fluorescence of other organs also could be observed at the 1st week. It was strong and wide in the liver, spleen and heart, while faint and less in the kidney and brain, and al reduced with time. It shows bone marrow mesenchymal stem cels transplantedvia the trachea into rats exposed to silica dust can be homing to the injured lung of rats.

ABSTRACT

AIM: To evaluate the protective effect of luteolin on endothelial dysfunction induced by tert-butyl hydro-peroxide (t-BOOH). METHODS: We observed the effect of luteolin on t-BOOH-induced contractions in the aorta rings with or without endot helium, which were incubated with luteolin (10-6 to 10-4 mol/L) for 30 min before determining the concentration-response to t- BOOH. Cultured endothelial cell line (ECV304) was pretreated with different concentrations of luteolin (10-6 to 10-4 mol/L) for 30 min and then exposed to 10 -5 mol/L t-BOOH for 24 hours. Cell morphology was observed , and cell viability was determined by MTT assay. Meanwhile, RT-PCR was used to measure the expression of eNOS and COX-1. RESULTS: Increasing concentrations of t-BOOH produced concentration-dependent cont ractions in aorta rings isolated from rats, luteolin effectively attenuated the contraction in a concentration-dependent manner, and the relaxation response was greater in intact endothelium segments. In MTT and RT-PCR assays, luteolin effectively reduced the cytotoxicity of t-BOOH to endothelium cell and increased the expression of nitric oxide synthase (eNOS) mRNA, which was greatly down-regulated by t-BOOH. CONCLUSION: Luteolin effectively protects the endothelium from the impairment of oxidative stress, and the protection could be related to its negative modulation towards t-BOOH-induced contractions in the aorta.
